Cytochrome P-450 catalysis: radical intermediates and dehydrogenation reactions

Abstract

Cytochrome P-450 appears to catalyse most monooxygenation reactions by sequential one-electron steps rather than by a single, concerted transfer of the ferryl oxygen to the substrate. The radical intermediates, although very short-lived, can rearrange or undergo alternative reactions characteristic of radical species. As Paul Ortiz de Montellano explains, these alternative reactions yield novel metabolites, sometimes result in inactivation of the enzyme, and can provide information on the reaction mechanism.

      Binding of oxazolinyl derivatives of [17(20)E]-pregna-5,17(20)-diene 1 and 2 to human recombinant CYP17A1 [24] was evaluated by absorption spectroscopy, as reported earlier for the characterization of interaction of steroidal CYP17A1 inhibitors abiraterone and TOK-001 with truncated, His-tagged version of the human CYP17A1 protein [21]. The spectrum of CYP17A1 (Fig. 2, insert) showed the maximum at 417 nm; titration of enzyme with substrate pregnenolone revealed absorbance decrease at 417 nm and increase at 386 nm, indicative of ligand displacing water from the heme (type I interaction [29,30]); titration of enzyme with abiraterone 3 caused appearance of characteristic maximum at 424 nm and minimum at 390 nm, corresponding to formation of coordinate bond between abiraterone pyridine nitrogen and heme iron (type II interaction [29,30]). Differential absorption spectra of CYP17A1 in the presence of pregnenolone, compounds 1, 2, and 3 recorded at saturating concentrations of ligands are shown in Fig. 2.
