Therapeutic relevance of pharmacokinetics and pharmacodynamics

Abstract

Most antineoplastic drugs have a narrow therapeutic index. The medical world's incomplete understanding of the variability of pharmacokinetics and pharmacodynamics has led to dosing problems with these agents. Basic understanding of linear and nonlinear pharmacokinetic models as well as interpatient and intrapatient variability is necessary to optimize dosing. Pharmacodynamic information relates pharmacokinetic information to clinical outcome.
