While this manuscript was undergoing review, Chen et al. (2005) published results demonstrating a 200% increase in Mrp4 protein expression in the kidney of TR- rats when compared to wild-type controls. Based on experiments conducted in our laboratory, the polyclonal Mrp4 antibody detailed by Leggas et al. (2004) provided similar results to a commercially available monoclonal antibody to MRP4 (M4I-10, Alexis Biochemicals, San Diego, CA). Our laboratory then repeated Mrp4 Western blot experiments in kidney tissue of TR- and wild-type rats (n=3 per group) using the polyclonal Mrp4 antibody detailed by Leggas et al. (2004). Mrp4 protein expression in TR- rats was slightly higher than that observed in wild-type controls; however, expression levels were not statistically different when normalized for b-actin. The apparent discrepancy in these results may be due to differences in experimental methodology and/or housing/dietary factors.
