P-gp inducers DEX and RIF (Sérée et al., 1998) decreased RhB and Rh123 accumulation, which indicates that they induced the efflux pump mechanism, i.e., enhanced the removal of fluorescent substrates from the cells, thereby decreasing fluorescence. The same induction effect was observed, for example, in earthworm Eisenia andrei exposed to DEX (Hackenberger et al., 2012) and porcine brain capillary endothelial cells treated with RIF (Ott et al., 2009).

Another interesting aspect concerning P-gp induction is the potential existence of a co-regulation mechanism of both P-gp and CYP3A4 expression (Sérée et al., 1998). As previously mentioned, P-gp is expressed in epithelial cells from tissues with secretory or excretory functions, as well as in endothelial cells of capillaries at blood-tissue barriers, having therefore a major influence in the absorption, distribution, metabolism and excretion of xenobiotics (Fojo et al., 1987; Thiebaut et al., 1987; Arceci et al., 1988; Chin et al., 1989; Cordon-Cardo et al., 1990; Saito et al., 1997; Lankas et al., 1998; Schinkel, 1999).
