Methotrexate is subject to liver metabolism with the formation of 7-hydroxymethotrexate under aldehyde oxidase action (Breithaupt and K ü enzlen, 1982; Moriyasu et al., 2006). Using partially purified aldehyde oxidase from rabbit liver (Johns et al., 1966; Johns and Loo, 1967), the 7-hydroxymethotrexate was obtained as the sole metabolite as expected (Fig. S1).
